Ah, okay, I see where the confusion is now.

So here's the key:

The Audio Device Assignment available in the preferences is only  
doing one thing: matching a patch number to an entire audio device.   
So, usually that means that you're telling QLab "Okay, I want all  
cues on patch number 3 to send their audio to my EMI.  All the cues  
on patch number 2 will send their audio to my internal speakers."   
And so on.  In other words, the patch assignment is not related to  
the individual channels on the device, but only to entire devices.

The place to adjust the routing for each channel on the device is in  
the "Levels" tab of the inspector.  That is where you can direct  
input from each channel of the file to each output channel of the  
device.
